Clioquinol or Iodochlorhydroxyquin is an antifungal drug and antiprotozoal drug. It is neurotoxic in large doses. It is a member of a family of drugs called hydroxyquinolines which inhibit certain enzymes related to DNA replication. The drugs have been found to have activity against both viral and protozoal infections.

DEFINITION
5-Chloro-7-iodoquinolin-8-ol.
Content: 98.0 per cent to 102.0 per cent (dried substance).

CHARACTERS
Appearance: Almost white, light yellow, brownish-yellow or yellowish-grey powder.
Solubility: Practically insoluble in water, sparingly soluble in methylene chloride, very slightly soluble or slightly soluble in ethanol (96 per cent).

A. Dissolve 40.0 mg in methanol and dilute to 100.0 mL with the same solvent. Dilute 10.0 mL to 100.0 mL with methanol (solution A). Examined between 280 nm and 350 nm, solution A shows an absorption maximum at 321 nm. Dilute 10.0 mL of solution A to 100.0 mL with methanol (solution B). Examined between 230 nm and 280 nm, solution B shows an absorption maximum at 255 nm. The specific absorbance at this absorption maximum is 1530 to 1660.
B. Infrared absorption spectrophotometry.
C. When heated, violet fumes are produced.
D. Dissolve about 1 mg in 5 mL of ethanol (96 per cent). Add 0.05 mL of ferric chloride solution. A dark green colour develops.

TESTS
Acidity or alkalinity: Shake 0.5 g with 10 mL of carbon dioxide-free water and filter. To the filtrate add 0.2 mL of phenolphthalein solution. The solution is colourless. Not more than 0.5 mL of 0.01 M sodium hydroxide is required to change the colour of the indicator to pink.

Loss on drying: Maximum 0.5 per cent, determined on 1.000 g by drying over diphosphorus pentoxide at a pressure not exceeding 0.7 kPa for 24 h.

ASSAY
Dissolve 0.200 g in 20 mL of acetic anhydride and add 30 mL of glacial acetic acid. Titrate with 0.1 M perchloric acid, determining the end-point potentiometrically. 1 mL of 0.1 M perchloric acid is equivalent to 30.55 mg of total quinolines, calculated as clioquinol.

Clioquinol, dried over phosphorus pentoxide for 5 hours, contains not less than 93.0 percent and not more than 100.5 percent of C9H5ClINO (the 5-chloro-7-iodo-8-quinolinol isomer).

Identification:
A: Chromatogram to pass the test.
B: Ultraviolet Absorption: To pass the test.
C: Heat 100 mg with 5 mL of sulfuric acid: copious violet vapors of iodine are evolved.
Loss on drying: Dry it over phosphorus pentoxide for 5 hours: it loses not more than 0.5% of its weight.
Residue on ignition: not more than 0.5%.
Free iodine and iodide: Shake 1.0 g with 20 mL of water for 30 seconds, allow to stand for 5 minutes, and filter. To 10 mL of the filtrate add 1 mL of 2 N sulfuric acid, then add 2 mL of chloroform, and shake: no violet color appears in the chloroform (free iodine). To the mixture add 5 mL of 2 N sulfuric acid and 1 mL of potassium dichromate, and shake for 15 seconds: the color in the chloroform layer is no deeper than that produced in a control test made in the following manner: Dilute 2.0 mL of potassium iodide solution (1 in 6000) with water to 10 mL, add 6 mL of 2 N sulfuric acid, 1 mL of potassium dichromate, and 2 mL of chloroform, and shake for 15 seconds (0.05% of iodide).
