Vanillin

Vanillin is the primary chemical component of the extract of the vanilla bean. Synthetic vanillin is an aromatic compound, Vanillin, as its name implies, is the major flavor component of vanilla. It is mostly used in foods, beverages, perfumes and pharmaceuticals.
Vanillin and ethylvanillin are used by the food industry; ethylvanillin is more expensive but has a stronger note. It differs from vanillin by having an ethoxy group (−O−CH2CH3) instead of a methoxy group (−O−CH3). Vanillin can be used as a general-purpose stain for visualizing spots on thin-layer chromatography plates.

Vanillin contains not less than 97.0 percent and not more than 103.0 percent of C8H8O3, calculated on the dried basis.

Melting range: between 81 and 83 .
Loss on drying: Dry it over silica gel for 4 hours: it loses not more than 1.0% of its weight.
Residue on ignition: not more than 0.05%.
Assay: Transfer about 100 mg of Vanillin, accurately weighed, to a 250-mL volumetric flask, add methanol to volume, and mix. Pipet 2 mL of this solution into a 100-mL volumetric flask, add methanol to volume, and mix. Dissolve an accurately weighed quantity of USP Vanillin RS in methanol and dilute quantitatively and stepwise with methanol to obtain a Standard solution having a known concentration of about 8 microg per mL. Concomitantly determine the absorbances of both solutions in 1-cm cells at the wavelength of maximum absorbance at about 308 nm, with a suitable spectrophotometer, using methanol as the blank. Calculate the quantity, in mg, of C8H8O3 in the portion of Vanillin taken by the formula:

in which C is the concentration, in microg per mL, of USP Vanillin RS in the Standard solution; and AU and AS are the absorbances of the solution of Vanillin and the Standard solution, respectively.

DEFINITION
Vanillin contains not less than 99.0 per cent and not more than the equivalent of 101.0 per cent of 4-hydroxy-3-methoxybenzaldehyde, calculated with reference to the dried substance.

CHARACTERS
White or slightly yellowish, crystalline powder or needles, slightly soluble in water, freely soluble in alcohol and in methanol. It dissolves in dilute solutions of alkali hydroxides.

A. Melting point: 81C to 84C.
B. Examine by infrared absorption spectrophotometry, comparing with the spectrum obtained with vanillin CRS. Examine the substances prepared as discs.
C. Examine the chromatograms obtained in the test for related substances in daylight after spraying. The principal spot in the chromatogram obtained with test solution (b) is similar in position, colour and size to the principal spot in the chromatogram obtained with reference solution (a).
D. To 5 mL of a saturated solution of the substance to be examined add 0.2 mL of ferric chloride solution. A blue colour is produced. Heat to 80C. The solution becomes brown. On cooling, a white precipitate is formed.

TESTS
Appearance of solution: Dissolve 1.0 g in alcohol and dilute to 20 mL with the same solvent. The solution is clear and not more intensely coloured than reference solution B.
Related substances: To pass the chromatographic test.
Any spot in the chromatogram obtained with test solution (a), apart from the principal spot, is not more intense than the spot in the chromatogram obtained with reference solution (b) (0.5 per cent).
Reaction with sulfuric acid: Dissolve 50 mg in 5 mL of sulfuric acid. After 5 min, the solution is not more intensely coloured than a mixture of 4.9 mL of yellow primary solution and 0.1 mL of red primary solution or a mixture of 4.9 mL of yellow primary solution and 0.1 mL of blue primary solution.
Loss on drying: Not more than 1.0 per cent, determined on 1.000 g by drying in a desiccator for 4 h.
Sulfated ash: Not more than 0.05 per cent, determined on 2.0 g.

ASSAY
Dissolve 0.120 g in 20 mL of alcohol and add 60 mL of carbon dioxide-free water. Titrate with 0.1 M sodium hydroxide, determining the end-point potentiometrically.
1 mL of 0.1 M sodium hydroxide is equivalent to 15.21 mg of C8H8O3.
