Sodium calcium edetate or Sodium calcium EDTA or Sodium Calcium Edetate or , also known as edetate calcium disodium among other names, is used as a medication to treat lead poisoning. It is a chelating agent, meaning that it binds to metals. When the metals are bound (or sequestered), they are prevented from taking part in chemical reactions that would lead to colour or flavor deterioration. Calcium disodium EDTA is a common popular food additive and flavoring agent and sequestrant, an ingredient in cosmetic and industrial products. It’s used in food to preserve flavor, color and texture. Calcium disodium EDTA is an odorless crystalline powder with a slightly salty flavor.

The USA FDA has approved calcium disodium EDTA as a safe food additive in small quantities. The E number of Calcium disodium EDTA is 385.

Calciate(2-), [[N,N′-1,2-ethanediylbis[N-(carboxymethyl)glycinato]](4-)-N,N′,O,O′,ON,ON′]-, disodium, hydrate, (OC-6-21)-. Disodium[(ethylenedinitrilo)tetraacetato]calciate(2-)hydrate [CAS 23411-34-9]. Anhydrous CAS [62-33-9].

Edetate Calcium Disodium is a mixture of the dihydrate and trihydrate of calcium disodium ethylenediaminetetraacetate (predominantly the dihydrate). It contains not less than 97.0 percent and not more than 102.0 percent of C10H12CaN2Na2O8, calculated on the anhydrous basis.
Packaging and storage: Preserve in tight containers.

Identification:
A: Infrared Absorption.
B: A solution (1 in 20) responds to the oxalate test for Calcium and to the flame test for Sodium.
C: To 5 mL of water add 2 drops of ammonium thiocyanate and 2 drops of ferric chloride. To the deep red solution add about 50 mg of Edetate Calcium Disodium, and mix: the deep red color disappears.
pH: between 6.5 and 8.0, in a solution (1 in 5).
Water: not more than 13.0%.
Heavy metals: 0.002%.
Limit of nitrilotriacetic acid: To pass the test (0.1%).
Magnesium-chelating substances: Weigh accurately 1.0 g, transfer to a small beaker, and dissolve in 5 mL of water. Add 5 mL of ammonia-ammonium chloride buffer. Then to the buffered solution add 5 drops of eriochrome black, and titrate with 0.10 M magnesium acetate to the appearance of a deep wine-red color: not more than 2.0 mL is required.
Assay: Weigh accurately about 1.2 g of Edetate Calcium Disodium, transfer to a 250-mL beaker, and dissolve in 75 mL of water. Add 25 mL of 1 N acetic acid and 1 mL of diphenylcarbazone, and titrate slowly with 0.1 M mercuric nitrate to the appearance of the first purplish color. Perform a blank determination, and make any necessary correction. Each mL of 0.1 M mercuric nitrate is equivalent to 37.43 mg of C10H12CaN2Na2O8.

DEFINITION
Disodium [(ethylenedinitrilo)tetraacetato]calciate(2-).
Content: 98.0 per cent to 102.0 per cent (anhydrous substance).
It contains a variable quantity of water of crystallisation.

CHARACTERS
Appearance: White or almost white, hygroscopic powder.
Solubility: Freely soluble in water, practically insoluble in ethanol (96 per cent).

IDENTIFICATION

A. Infrared absorption spectrophotometry.
B. Dissolve 2 g in 10& mL of water, add 6 mL of lead nitrate solution, shake and add 3 mL of potassium iodide solution. No yellow precipitate is formed. Make alkaline to red litmus paper by the addition of dilute ammonia and add 3 mL of ammonium oxalate solution. A white precipitate is formed.
C. Ignite. The residue gives a reaction of calcium.
D. Dissolve 0.5 g in 10 mL of water and add 10 mL of potassium pyroantimonate solution. A white, crystalline precipitate is formed. The formation of the precipitate is accelerated by rubbing the wall of the tube with a glass rod.

TESTS
Solution S: Dissolve 5.0 g in water and dilute to 100 mL with the same solvent.
Appearance of solution: Solution S is clear and colourless
pH: 6.5 to 8.0.
Dissolve 5.0 g in carbon dioxide-free water and dilute to 25 mL with the same solvent.
Impurity A (nitrilotriacetic acid): To pass the test by liquid chromatograph.

DESCRIPTION
Calcium Disodium EDTA occurs as white, crystalline granules or as a white to off white powder. It is slightly hygroscopic and is stable in air. It is freely soluble in water.
Function: Preservative; sequestrant.

REQUIREMENTS
Identification:
A. A 1:20 aqueous solution responds to the oxalate test for Calcium and to the flame test for Sodium.
B. The infrared absorption spectrum of a mineral oil dispersion of sample exhibits maxima only at the same wavelengths as those of a similar preparation of USP Edetate Calcium Disodium Reference Standard.
C. Add 2 drops of ammonium thiocyanate and 2 drops of ferric chloride to 5 mL of water contained in a test tube. Add about 50 mg of sample to the deep red solution so obtained, and mix. The deep red color disappears.
Assay: Not less than 97.0% and not more than 102.0% of C10H12CaN2Na2O8, calculated on the anhydrous basis.
Lead: Not more than 4 mg/kg.
Magnesium-Chelating Substances: Passes test.
Nitrilotriacetic Acid: Not more than 0.1%.
pH of a 1:100 Solution: Between 6.5 and 7.5.
