Neomycin Sulfate

Neomycin sulfate is an aminoglycoside antibiotic and calcium channel protein inhibitor. Neomycin sulfate also binds to prokaryotic ribosomes inhibiting translation and is effective against gram-positive and gram-negative bacteria.

Neomycin Sulfate is the sulfate salt of a kind of neomycin, an antibacterial substance produced by the growth of Streptomyces fradiae aksman (Fam. Streptomycetaceae), or a mixture of two or more such salts. It has a potency equivalent to not less than 600 µg of neomycin per mg, calculated on the dried basis.
Packaging and storage: Preserve in tight, light-resistant containers.

Identification:
A: It meets the requirements for neomycin under Thin-Layer Chromatographic Identification Test.
B: Dissolve about 10 mg in 1 mL of water, add 5 mL of 15 N sulfuric acid, and heat at 100C for 100 minutes. Allow to cool, add 10 mL of xylene, and shake for 10 minutes. Allow to separate and decant the xylene layer. To the xylene layer add 10 mL of p-bromoaniline TS, and shake: a vivid pink-red color develops upon standing.
C: A solution (1 in 20) responds to the tests for Sulfate.

pH: between 5.0 and 7.5, in a solution containing 33 mg of neomycin per mL.
Loss on drying: Dry about 100 mg in vacuum at a pressure not exceeding 5 mm of mercury at 60C for 3 hours: it loses not more than 8.0% of its weight.
Other requirements: Where the label states that Neomycin Sulfate is sterile, it meets the requirements for Sterility and Bacterial endotoxins under Neomycin for Injection.

DEFINITION
Mixture of sulfates of substances produced by the growth of certain selected strains of Streptomyces fradiae, the main component being the sulfate of 2-deoxy-4-O-(2,6-diamino-2,6-dideoxy-α-D-glucopyranosyl)-5-O-[3-O-(2,6-diamino-2,6-dideoxy-β-L-idopyranosyl)-β-D-ribofuranosyl]-D-streptamine (neomycin B).
Content: Minimum of 680 IU/mg (dried substance).

CHARACTERS
Appearance: White or yellowish-white powder, hygroscopic.
Solubility: Very soluble in water, very slightly soluble in alcohol, practically insoluble in acetone.

4. First Aid Measures

Always seek medical attention after first aid measures are provided.

Eye Contact: Wash out immediately with fresh running water. Ensure complete irrigation of the eye by keeping eyelids apart and away from eye and moving the eyelids by occasionally lifting the upper and lower lids. Seek medical attention without delay; if pain persists or recurs seek medical attention. Removal of contact lenses after an eye injury should only be undertaken by skilled personnel.
Skin Contact: Immediately remove all contaminated clothing, including footwear. Flush skin and hair with running water (and soap if available). Seek medical attention in event of irritation.
Inhalation: Lay patient down. Keep warm and in rest. Prostheses such as false teeth, which may block airway, should be removed, where possible, prior to initiating first aid procedures. Apply artificial respiration if not breathing, preferably with a demand valve resuscitator, bag-valve mask device, or pocket mask as trained. Perform CPR if necessary. Transport to hospital, or doctor, without delay.
Ingestion: If swallowed do NOT induce vomiting. If vomiting occurs, lean patient forward or place on left side (head-down position, if possible) to maintain open airway and prevent aspiration. Observe the patient carefully. Never give liquid to a person showing signs of being sleepy or with reduced awareness i.e., becoming unconscious. Give water to rinse out mouth, then provide liquid slowly and as much as casualty can comfortably drink. Seek medical advice.
